In my laboratory we studied the domestic chicken (Gallus domesticus). The chicken is an important source of protein (meat and eggs) for people around the globe. Research on the chicken is important because of its agricultural significance and because it serves as an excellent animal model in many fields, e.g., for human diseases, developmental biology and comparative vertebrate biology.

Students studying in the lab (M.S. and Ph.D.) were trained in the disciplines of genetics, cytogenetics, and genomics with an emphasis on avian systems as well as comparative vertebrate biology. Research and technology levels ranged from molecular and cellular to the organismal.

Chromosome Organization.

In order to explore and better understand chromosome organization of chicken, turkey and quail we employed molecular cytogenetics to study chromosome architecture, gene and gene complex order, and the mapping of coding and repetitive non-coding elements. Studies included in depth analysis of individual chromosomes (e.g, GGA16 and GGA17) to comparative analysis among poultry genomes and specialized resources (in vitro cell lines).

Telomere Biology

Telomere stability is one of the most significant genetic mechanisms controlling overall genome stability and influencing cellular proliferation, senescence and transformation. Our work focused on the organization, inheritance, regulation and stability of telomere array organization in normal, immortalized and transformed cell systems of the chicken.

Host-Herpesvirus Genome Interactions

Projects included the regulation and function of the telomere-telomerase pathway during transformation and tumorigenesis caused by Marek’s disease virus (MDV), a DNA herpes virus which induces T-cell lymphomas and results in a high level of mortality. This particular disease is a problem of enormous significance for the poultry industry. MDV infection and disease in chickens also serves as a model system for human herpes virus infection and disease conditions (e.g., Burkitt’s lymphoma caused by Epstein Barr virus). We determined that the MDV integrates into the telomeres of the chicken genome randomly and ubiquitously during early infection and then during latency and oncogenesis wherein the further selective mechanisms streamline those lineages leading to the oncogenic tumorigenic condition of the disease.

Developmental Mutations

Another main research area included the mapping, sequencing and analysis of priority candidate regions to discover the elements causative of inherited mutations of the face (e.g., cleft palate) and limbs (wingless). Such studies contributed new knowledge of key developmental pathways in chicken and serve as models for human development and comparative vertebrate biology.

Conservation of Research Genetic Resources

All of our work employed specialized genetic resources (inbred and congenic inbred lines of chicken (living stocks); in vitro cell lines) that were developed by geneticists working with poultry since the 1950s and maintained at UC Davis, Cornell University (Ithaca, NY) and the USDA-ARS Avian Disease and Oncology Laboratory (E. Lansing, MI). Prior researchers maintained, characterized these lines as well as explored important biological questions relative to poultry and vertebrate biological mechanisms. We were able to further the state of fundamental knowledge in the areas outlined above utilizing these important resources. A key issue that has yet to be resolved is how to maintain such irreplaceable resources in the face of financial issues and staffing/space pressures for Agricultural Experiment Stations at the land grant institutions.
